Abstract
I denne oppgaven ønsket vi å studere celleoverlevelse, definert som evne til kolonidannelse, hos brystkreftlinjen T-47D etter behandling med autofagihemmer under hypoksiske (4 % O2) og normoksiske (21 % O2) forhold. Autofagihemmerne 3-methyladenine og chloroquine ble benyttet, og målet var å finne den høyeste konsentrasjonen cellene kunne dyrkes med uten å redusere overlevelsesfraksjonen. Det er tidligere beskrevet en omvendt oksygeneffekt med høyere dødelighet og større tap av kolonidannende evner hos T-47D celler ved høy (19 %) kontra lav (4 %) O2-metning i gassfase (Pettersen et al. 2007). Det har blitt spekulert i om man ved å hemme autofagi kan øke denne effekten ytterligere. Denne oppgavens formål var således å finne en egnet autofagihemmer til videre forsøk, samt bestemme hvilken konsentrasjon en helst bør benytte. I tillegg ble det gjort et pilotforsøk der effekten av autofagihemmer i dyrkningsmediet kombinert med lavdoseratebestråling i hypoksi ble undersøkt.
Reduced oxygen levels, hypoxia, is a common feature of solid tumors, and has been shown to reduce therapy outcome and overall survival. Under hypoxia and other stressful conditions, cells activate autophagy, which is a basic cellular process that allows the cells to survive by recycling its own components. Increasing evidence suggests that activation of autophagy plays a crucial role in the resistance of hypoxic cancer cells to therapy, and there is need for more knowledge about the consequences of autophagy modulation. We wanted to investigate if addition of autophagy inhibitors affected the survival of the breast cancer cell line T47-D under normoxic (21 % O2) and hypoxic (4 % O2) growth conditions. The aim was to find the highest concentration of autophagy inhibitors 3-methyladenine (3-MA) and chloroquine (CQ) that could be added to the growth medium without affecting the survival fraction. We also wanted to investigate whether addition of autophagy inhibitor to the growth medium could increase the effect of low dose rate irradiation under hypoxic condition. We found that CQ had unspecific toxic effects on the cells in concentrations from 10 µM to 10 mM under normoxic conditions. The cells tolerated 3-MA in concentrations from 1 µM to 1 mM well, both under normoxic and hypoxic conditions. Five mM had slight toxic effects under hypoxic conditions. Interestingly, we also found indication of effect by combining the addition of 5mM 3MA to the growth medium with irradiation by addition of [3H]Val to the growth medium.
