Abstract
EPO WGA MAIIA Isoformdistribution Kit er en ny metode for deteksjon av rekombinant og endogent EPO i blod- og urinprøver. Metoden er en kombinasjon av en kromatografisk separasjonsmetode og et sandwich immunoassay for deteksjon. Metoden er konstruert slik at både separasjon og deteksjon skjer på en tynn porøs membranstrips ved hjelp av lateral flow gjennom stripsmembranen.
Metoden viser stort potensial i forhold til eksisterende metoder som IEF- og SDS-/SAR-PAGE. Den kan skille rekombinante EPO varianter i både urin- og blodprøver. Samtidig har den også mye lavere deteksjonsgrenser i forhold til eksisterende metoder som gjør at deteksjonsvinduet til rhEPO kan forlenges til det dobbelte. På grunn av sin lave deteksjonsgrense gir det også mulighet til å fange intravenøse injeksjoner av mikrodoser rhEPO.
Den utførte studie av mikrodosen injeksjoner (15 IU/kg/uke) av rhEPO (NeoRecormon) bekrefter disse påstandene med prøveresultater som viser signifikante forskjeller i prøver tatt opp til 24- timer etter intravenøs mikrodose.
EPO WGA Maiia Isoformdistribution Kit is a new method for the detection of recombinant and endogenous EPO in blood and urine samples. The method is a combination of a chromatographic separation method and a sandwich immunoassay for detection. The method is designed so that both the separation and detection takes place on a thin porous membrane strips using lateral flow through the membrane strips.
The method shows great potential as compared to existing methods IEF and SDS-/SAR-PAGE. It can distinguish recombinant EPO variants in both urine and blood samples. At the same time, it also has much lower detection limits compared to existing methods that allow the detection window of rhEpo can be extended to twice. Because of its low detection limit, it also provides the ability to capture intravenous injections of micro doses rhEpo.
The study carried out by micro-dose injections (15 IU / kg / week) of rhEpo (NeoRecoromon) confirms these claims with test results showing significant differences in samples taken up to 24 - hours after intravenous micro dose.
