dc.description.abstract | Bakgrunn:
Det er sterke bevis på at synaptisk plastisitet kan spille en rolle i patiofysologien til og behandlingen av både unipolar og bipolar depressiv lidelse. Men det meste av disse bevisene er indirekte, enten fra dyremodeller eller in vitro studier, grunnet mangel på metoder for å måle synaptisk plastisitet non-invasivt. Men nylig har det blitt vist at det er mulig å indusere VEP-plastisitet gjennom repetitiv visuell stimulering, og denne plastisiteten tror man gjenspeiler synaptisk plastisitet i synsbarken.
Metode: 40 friske kontroller og 26 pasienter med bipolar lidelse type 2 (BD-II) ble rekruttert. Gruppene var like med tanke på alder og kjønn. VEP-plastisitet ble indusert ved at forsøkspersonene så på et sjakkbrettmønster i 10 minutter som endret seg fra hvitt til sort og omvendt to ganger i sekundet, og målt ved å måle VEP-amplitudene før og etter denne perioden.
Resultater:
Vi fant signifikant VEP-plastisitet hos friske kontroller. Motsatt fant vi ingen signifikant VEP-plastisitet hos personer med BD-II. Videre var forskjellen i plastisitet mellom friske kontroller og bipolare signifikant. Videre analyse viste at medisinerte bipolare kunne ha mindre skadet plastisitet en umedisinerte, men trenden var ikke signifikant.
Konklusjon:
Vi reproduserte tidligere funn av VEP-plastisitet hos friske kontroller, noe som indikerer at dette er en robust og pålitelig måte å måle synaptisk plastisitet på in vivo hos mennesker. Videre støtter våre funn at skadet synaptisk plastisitet spiller en rolle ved bipolar lidelse type 2. Videre longitudielle studier trengs for å grundigere klargjøre våre funn, og får å se hvor stor innvirkning medikamentering og humør har på VEP-plastisitet. | nor |
dc.description.abstract | Background: There is ample evidence that synaptic plasticity might play an important role in the pathophysiology and treatment of both unipolar and bipolar depressive disorders. Most of this evidence, however, is indirect through animal and in vitro studies, due to lack of methods to assess synaptic plasticity noninvasively. Recently, though, it has been shown that it is possible to induce plasticity of the VEP by repeated visual stimulation, and this plasticity is believed to reflect synaptic plasticity in the visual cortex. In this study, we examined the plasticity of the VEP in healthy controls and compared it to that in patients with bipolar disorder II (BD-II).
Methods: 40 healthy controls and 26 patients with BD-II were recruited. The two groups were matched for gender and age. Plasticity of the VEP were induced by a ten minutes checkerboard reversal period, and measured by inducing VEPs before and after this period.
Results: We found significant VEP plasticity in the healthy control subjects. Conversely, we did not find any significant VEP plasticity in subjects with BD-II. Also, the difference between healthy controls and BD-IIs were significant. Further analyses also indicated that medicated subjects with BD-II had a less severely impaired plasticity of the VEP than unmedicated, although the trend was not significant.
Conclusion: We reproduced previous findings of plasticity of the VEP in healthy control subjects, suggesting that this is a reliable and robust way to assess synaptic plasticity in vivo in humans. Furthermore, our findings support the hypothesis that impaired synaptic plasticity plays a part in BD-II. We conclude that future longitudinal studies are required in order to further clarify our findings and further explore the extent to which medication and mood state affects plasticity of the VEP. | eng |
