Abstract
En metode for bestemmelse av 99Tc i urin og serum med ICP-SF-MS er blitt utviklet og validert.
Metoden muliggjør bestemmelse ned i konsentrasjoner på under 10 ng/l i urin og serum. I medisinsk diagnostikk brukes den radioaktive isotopen 99mTc. Den nye metoden detekterer 99Tc slik at man ikke er avhengig av den korte halveringstiden til isotopen 99mTc og heller ikke trenger tenke på radioaktiv stråling.
De ulike betingelsene for instrumentet er systematisk vurdert og optimalisert for å oppnå best mulig sensitivitet ved kortest mulig analysetid.
Metoden ble benyttet for å bestemme Tc i blod og urin fra rotter som har fått injisert 99mTc-DTPA intravenøst. Blod- og urinprøvene ble også målt med tradisjonell gammatelling på 99mTc for sammenlikning. Det er vist at de to metodene gir tilsvarende resultater.
A method for quantification of 99Tc in biological material with high resolution ICP-SF-MS has been developed and validated.
The method makes it possible to determine Tc at levels of below 10 ng/l in urine and serum. The radioactive isotope 99mTc is often used diagnostically. The new method detects 99Tc therefore one does not have to take notice of the short half-life of the isotope 99mTc, nor the radioactive radiation.
The variables of the instrument have been systematically optimized to gain the highest sensitivity as well as the shortest analytical duration as possible.
The method has been used to determine Tc in blood- and urine-samples from rats that have been intravenous injected 99mTc-DTPA. The blood- and urine-samples have also been analyzed for 99mTc by traditionally gamma counting. It is showed that the two methods give equivalent results.
